Medicine overview

Danogen-200 is an oral prescription medicine containing danazol 200 mg. Although the supplied product name describes it as a tablet, regulatory product records identify Danogen-200 as a capsule manufactured by Cipla Ltd. Danazol is a synthetic steroid with weak androgenic activity that suppresses parts of the hormonal system controlling ovarian function. It is used for conditions including endometriosis and, where clinically appropriate, severe fibrocystic breast disease and prevention of hereditary angioedema attacks.

What it is used for

Danazol is used to treat endometriosis that is suitable for hormonal treatment. It may also be used for severe pain and tenderness associated with fibrocystic breast disease when simpler treatments have not provided sufficient relief. Another established use is preventing attacks of hereditary angioedema, an inherited condition that can cause episodes of swelling affecting areas such as the face, limbs, abdomen, or airway.

How it works

Danazol suppresses the pituitary-ovarian hormonal axis and reduces the release of follicle-stimulating hormone (FSH) and luteinizing hormone (LH). In endometriosis, these hormonal effects make endometrial tissue less active and promote its shrinkage. Danazol also has weak androgen-like activity. In hereditary angioedema, it can increase levels of deficient C1 esterase inhibitor, helping reduce the occurrence of swelling attacks.

How it is taken

Danogen-200 is taken by mouth as a capsule. Take danazol exactly as prescribed and do not take more, less, or more frequently than directed. The required regimen varies substantially according to the condition being treated and the patient's response, so a patient-specific schedule should come from the prescriber. For endometriosis, treatment is generally started during menstruation; otherwise pregnancy should be excluded before treatment begins. Food can substantially increase danazol absorption, so follow the prescriber's instructions consistently regarding meals.

Important warnings

Danazol must not be used during pregnancy or breastfeeding. Contraindications include undiagnosed abnormal genital bleeding, markedly impaired liver, kidney or heart function, porphyria, androgen-dependent tumors, active or previous thrombosis or thromboembolic disease, and hypersensitivity to danazol. Serious reported risks include blood clots, stroke, liver injury and rare liver tumors with long-term use, and benign intracranial hypertension. Seek prompt medical assessment for severe or persistent headache with nausea, vomiting or visual changes, symptoms suggestive of a blood clot, or signs of liver problems. Some androgenic effects, including voice changes, may not completely reverse after stopping treatment. Important interactions include warfarin, carbamazepine, cyclosporine, tacrolimus, antidiabetic medicines, and some statins including simvastatin, atorvastatin and lovastatin.

Side effects and monitoring

Common or characteristic effects include weight gain, acne, oily skin, hair growth, hair loss, fluid retention, menstrual changes or absence of periods, flushing, sweating, vaginal dryness or irritation, and voice changes. Nausea, vomiting, headache, dizziness, fatigue and mood changes have also been reported. Serious or less common problems include jaundice or other liver dysfunction, thromboembolic events, severe skin reactions and benign intracranial hypertension. Periodic liver-function testing is recommended, and hematologic monitoring may be considered. Patients should also be observed for androgenic changes because some may persist after treatment ends.

Pregnancy

Contraindicated during pregnancy. Danazol can cause androgenic effects in a female fetus. Pregnancy should be excluded immediately before treatment, and a non-hormonal method of contraception should be used during therapy. If pregnancy occurs during treatment, the medicine requires prompt medical review.

Breastfeeding

Contraindicated during breastfeeding.

Food and drink

Food can substantially increase danazol exposure. In pharmacokinetic studies, a meal containing more than 30 g of fat increased danazol availability and peak blood concentrations compared with fasting. Follow the prescriber's instructions consistently regarding whether the medicine is taken with food.

Storage

Store below 30°C.

Frequently asked questions

How long does danazol take to start helping endometriosis symptoms?

Response varies between patients and the prescribed regimen. Danazol suppresses hormonal stimulation of endometrial tissue, but symptom improvement is not necessarily immediate; continue it only according to the prescribed treatment plan.

Can Danogen-200 change or stop my periods?

Yes. Danazol can cause spotting, changes in cycle timing, irregular periods or amenorrhea, meaning periods stop. Ovulation and cyclic bleeding usually return after treatment ends, although persistent amenorrhea has occasionally been reported.

Can danazol cause acne, hair changes or a deeper voice?

Yes. Danazol has androgen-like effects and can cause acne, oily skin, increased body hair, hair loss and voice changes. Some androgenic changes, particularly voice changes, may persist after the medicine is stopped.

Can danazol cause weight gain?

Yes. Weight gain and fluid retention have been reported during danazol treatment. Fluid retention is particularly relevant for people with conditions such as heart or kidney dysfunction, hypertension, migraine or epilepsy.

What should I do if I develop a severe headache or vision changes while taking danazol?

Seek prompt medical assessment. Danazol has been associated with benign intracranial hypertension; warning symptoms can include headache, nausea, vomiting and visual disturbances.

Can I take Danogen-200 during pregnancy?

No. Danazol is contraindicated during pregnancy. Pregnancy should be excluded before treatment, and non-hormonal contraception is recommended during therapy.

Does food affect Danogen-200?

Yes. Food, particularly a high-fat meal, can substantially increase the amount of danazol absorbed. Follow the prescriber's directions consistently regarding meals.

Is Danogen-200 a tablet or a capsule?

It is a capsule. Regulatory records identify Danogen-200 as a 200 mg danazol capsule manufactured by Cipla Ltd.; the supplied description as a “tablet” does not match the verified dosage form.
