Medicine overview

Danogen 50 mg is an oral capsule containing 50 mg of danazol, a synthetic steroid with androgenic and hormone-suppressing effects. The Danogen 50 mg product is manufactured by Cipla Limited. Danazol is used for hormone-responsive endometriosis and can also be used to prevent attacks of hereditary angioedema. Its hormonal effects can cause significant adverse reactions, so treatment requires medical supervision and appropriate monitoring.

What it is used for

Danazol is used to treat endometriosis that is suitable for hormonal treatment. By suppressing hormonal stimulation of endometrial tissue, it can reduce endometriosis-related symptoms. Danazol is also indicated for prevention of hereditary angioedema attacks, including swelling affecting the skin, abdomen, or airway. How it works

Danazol suppresses the pituitary-ovarian hormonal system and has weak androgenic activity. In endometriosis, its hormonal effects reduce stimulation of endometrial tissue, producing changes that can make endometriotic lesions inactive. In hereditary angioedema, danazol increases levels of deficient or reduced complement-related proteins, including C1 esterase inhibitor, which helps reduce susceptibility to swelling attacks.

How it is taken

Danogen is taken by mouth as a capsule. Use it exactly according to the prescribed regimen; the required amount and duration differ substantially according to the condition being treated and individual response. For endometriosis, treatment should not be started until pregnancy has been excluded. Food can substantially increase danazol absorption, particularly a meal containing more than 30 g of fat, so follow the prescriber's instructions about how to take each dose in relation to meals.

Important warnings

Danazol must not be used during pregnancy or breastfeeding. It is also contraindicated in people with unexplained abnormal genital bleeding, markedly impaired liver, kidney, or heart function, porphyria, androgen-dependent tumors, active or previous thrombosis or thromboembolic disease, or hypersensitivity to danazol. Serious reported risks include blood clots and stroke, severe liver injury, peliosis hepatis, hepatic adenoma, and benign intracranial hypertension. Some androgenic changes, particularly voice changes, may not fully reverse. Important interactions include warfarin, carbamazepine, cyclosporine, tacrolimus, antidiabetic medicines, and certain statins including simvastatin, atorvastatin, and lovastatin. Urgent assessment is needed for sudden chest pain or breathlessness, symptoms suggesting a blood clot or stroke, jaundice, or severe headache with vomiting or visual disturbance.

Side effects and monitoring

Common or characteristic effects include weight gain, acne, oily skin, mild increased body or facial hair, fluid retention, hair loss, flushing, sweating, vaginal dryness, menstrual changes, spotting, or absence of periods. Voice deepening or other voice changes can occur and may persist after treatment. Serious or less common effects include liver injury, jaundice, thrombosis, stroke, intracranial hypertension, severe skin reactions, and significant blood-count abnormalities. Periodic liver-function testing is recommended, and hematologic monitoring should be considered. Danazol can also alter cholesterol and other laboratory measurements. Seek prompt medical attention for jaundice, severe or persistent headache with visual changes, signs of thrombosis, severe allergic or skin reactions, or other significant new symptoms.

Pregnancy

Danazol is contraindicated during pregnancy because fetal exposure can produce androgenic effects, particularly masculinization of a female fetus. Pregnancy should be excluded before treatment begins, and a non-hormonal contraceptive method is recommended during therapy. If pregnancy occurs during treatment, medical advice should be obtained promptly.

Breastfeeding

Danazol is contraindicated during breastfeeding. Current evidence does not provide adequate information about danazol exposure through human milk, and an alternative medicine is preferred during lactation.

Food, drink and interactions

Avoid alcohol while taking danazol. Patient guidance reports that combining alcohol with danazol can cause nausea or shortness of breath.
